COMPARISON OF MICROSCOPIC, FLOW CYTOMETRIC AND COULTER STKS ANALYSER METHODS FOR RETICULOCYTE COUNTING

We compared microscopic (brilliant cresyl blue stain), flow cytometric (thiazole orange technic), coulter STKS (VCS technology) reticulocyte count methods. There were good correlations between microscopic and flow cytometric reticulocyte counts (r=0.94), microscopic and coulter STKS analyser reticulocyte counts (r=0.84), flow cytometric and coulter STKS analyser reticulocyte counts (r=0.83). Coefficient of variation (CV) were 14.6% for microscopic method. 9.7% for flow cytometric method, 11.8% for coulter STKS method. Our data suggests that flow cytometric and coulter STKS analyser reticulocyte count methods are reliabte for routine reticulocyte count.
