PROTECTION OF HEMATOPOIETIC PROGENITOR CELLS FROM CHEMOTHERAPY TOXICITY BY GENE TRANSFER

Several genes have been studied to render hematopoietic progenitor cells resistant to dose limiting toxicity from chemotherapy. This area of research will not only allow us to give high dose chemotherapy but to expand its use for a selectable marker with other gene of interest especially requiring higher expression of the gene to correct its underlying disease (e.g. § globin). The most important issue which limits the effectiveness of hematopoietic stem cell gene transfer, is low transduction efficiency. One promising way to rescue the hematopoietic cells from chemotherapy toxicity is retroviral gene transfer of drug resistance genes. This method and drug resistance genes have been reviewed in this paper.
